I've been researching for new boots on the Zook, and while the 215/75x15 seem fit a stock Sierra with no modification, the question is how much in reality do larger tyres affect driveability? I understand larger tyres affect acceleration, fuel economy, highway cornering etc, but as a daily drive with the odd offroad getaway, is upgrading from stock tyres to 215/75 profile going to make me wish I kept stock sizing?

No they will be fine - little size difference over standard. I've run that size on daily driven Sierras for several years.
